You know, for those of you trying to beat the first boss, you could just read the game instructions XD. I actually only beat it when I right-clicked by accident.

To beat the first boss, move your mouse over the left hand white box so that a ring appears around the button, then right-click and then click on the button.
To beat the second boss, there are three locks to open: one on the left, one on the right and one above the padlock. Click each of these and the button will appear.
To beat the last boss, wait until the arrow (not your cursor) is over the button, then click.
