Ive just unlocked gathering/crafting, and i was wondering the best way tk increase my reincarnation bonus. I have completed no challenges, will those help? if so, Which ones? I have inferno, Spiritus, and basic.

I'm not sure what would be "best", but...
A lot of this game is layer upon layer upon layer of different features feeding into each other. But ignoring that for a really basic surface level analysis, you can think of reincarnation as being pretty much just what it looks like from the reincarnation screen: highest stage & mana spent on conjuring & demon meditation. And without getting too complicated about it, making progress in all of those should pretty much be a matter of working on your mana cap, mana power, and also mana refill.
You need offense and defense to advance through the stages. Having more mana to invest in spells, and having more mana power to improve the effectiveness of the mana that you invest in the spells, improves your offense and defense so that you can push through to higher stages.
Each next level in each thing in conjuring costs more mana than the previous level did. The more mana you have to throw around, the more levels you'll be able to get and the more you'll have spent in total on them. Some of the conjuring options are also gated by needing to reach a high enough stage first.
Meditation levels are gained based on the number of demons you have working on any of the six buffs (or the generic one as well) on the demons screen. To get more demons you need conjuring levels, which ties back into the previous point. More mana -> more conjuring -> more demons -> more meditation.
Mana refill doesn't necessarily boost anything directly, I guess... But having more mana refill will mean getting quicker access to all of your mana to dump into spells and conjuring. Also, every time you generate mana, even when you're at your cap already, that counts towards a bonus for the next life's mana cap when you reincarnate. You'll gain a lot more cap this way than you will from spending exp on it, actually. (Honestly, cap purchased with exp will prove to be more relevant in challenges since those temporarily remove what you've built up and start you with just your purchased mana cap.
Oh, with crafting available to you... Well, one of the three gems you can make will give a bonus to meditation. So if you make and equip that it will improve the leveling speed of meditation a little bit. The respawn gem will affect respawn times for the stages, I think. (Unless it's talking about minion adventure enemy respawn? But if it's for stages that can mean better gold and exp earnings.) And the other gem you're initially able to make is for mana refill.
As for challenges, specifically the three you mentioned...
The first completion of the First Step challenge (the one you called "basic") unlocks the fourth dungeon. (Dungeons require shadows, which you get based on the amount of spirit you currently have. Spirit refill comes from the completion of the demon reproduction bar on the demons screen.) Completing levels in that dungeon boosts your gold and exp earnings that you get from the stages you're beating. Gold is used in the town screen (which has a whole bunch of different bonuses that you can level up, including things mentioned above in this post), and exp can be used to bulk up your mana stats.
The first completion of the Inferno challenge unlocks a couple stronger spells that you can use. They're slower than the spells you've been able to use so far, though. Requiring even more mana and mana power to be able to get a decent number of levels.
The first completion of the In Spirtus challenge unlocks the ninth dungeon. Completing levels in that one buffs your spirit refill speed. And the benefit in that for your reincarnation bonus.... Well, just like how generating mana builds up a bonus for mana cap that you'll receive when you reincarnate, generating spirit does the same thing for your spirit cap. (And there's no purchasing more spirit cap. It's only obtained in this way.) Spirit is used in the generator (which has bonuses for a ton of different things, including a direct buff to your reincarnation bonus) and to some extent also in gathering. If you see any bonus that you can pick up in the game which buffs "essence" that means that the output of the generator's bonuses (and the degree to which charges can increase the permanent part of the generator's bonuses) is buffed.
Moving on from those challenges... The first completion of the Demon Master challenge will unlock a dungeon that boosts meditation. The first completion of No Conjuring unlocks a dungeon that reduces conjuring costs (which can mean being able to afford more levels and spending more overall). The first completion of Time Loop unlocks a dungeon that directly buffs the reincarnation bonus.
