Level 45: Quickly expand your territory to accessible jellies, then send everything onto the defense jelly. Then, send the defense jelly toward the big bad boss, and proceed to throw everything at once at him before he gets to cast his first spell. If done well, you'll steal the big boss but lose all you had. Then just play smarter than him, stealing back is easy after this.
