Page 1 of 2: Glitch: Those are what that particular tower will target as priority bloons to attack. I will give you an example, if a tower is near the start and bloons are still entering the map and also passing out of range of the tower, but the majority are near it, your targeting of the tower affects how it attacks. If you choose First, it will attack those that are earliest in the map, those getting out of its range, trying to keep bloons from escaping it. If you choose Last, it will attack those first entering the map (the earliest part of the map where it can reach). If you choose Close, it will attack those which are nearest the tower. And if you choose Strong, it will attack the strongest bloons.
Page 2 of 2 (cont....): For myself, it depends on the location, the type of tower and its abilities, and the type of bloons attacking (is it earlier in the game, or much later, where there are more potent types of bloons), for me to make my selection. Near the start, with more potent bloons, I usually have a Bomb Tower attacking strong bloons to help break up difficult bloons for the other towers to deal with the remnants. At the start, I often have a Glue Gunner going after strong bloons to affect all the bloons inside and help slow them down . The Boomerang Thrower solely depends on its location and where it would be best to use its curve. Towers late in the map, I set to First, as I don't want any bloon to escape, etc. Choose carefully as it can make a huge difference.

MouseCheese4All: To close the Evil Graveyard Portal you simply need 15 soldiers, 3 defensive buildings, and 50% happiness, which will take 10 days to accomplish. If you have any further questions, feel free to pm me.

Selyob: All the orb rooms have a purpose. The yellow ones will heal a whole party [once], which you choose when to use. The blue ones vary from what XOXSarahXOX lists above. You didn't mention the green question marks, but those are either the Lurker requesting you to clean the floor for some items or an adventurer asks to join your party until they die [can not be revived].
Part of the strategy is for you to carefully choose which rooms to enter, so you may not wish to enter all at one time so that you can heal as you walk, when needed. For resting and healing there is both the yellow orb mentioned above, and you can [one time] retire to the prior level.
As for your items disappearing, I have no idea what you're talking about. You could pm me with a better description and I can try to help you.
Yes, there are Bosses which can revive others. You need to strategize how to best deal with that.

Two brief suggestions regarding strategies you may require: As for powerful bosses, my best option against them is using the Rogue's “Sap Power;” and as for Healing, my Ranger uses Nature's Balm on my people and I let them all heal and replenish power before finishing off the last of the enemies, so I have no need to heal while walking.
This is a good strategy game. You just must not have completely figured the game out yet, and perhaps have the wrong group suited to your personality. If you have any questions, feel free to pm me.
Oranges: If your people are killing themselves, then you have allowed the Church of the Chosen Ones. Next time, avoid it by not allowing them to write the book about the zombies in the first place. Many people do allow them to do that (for some needed happiness), but they do say NO to letting them set up the Church. This will solve that problem.
YT: Why are you tending the farms? Simply travel the direction with the most farms (and some living quarters), and reclaim as many of them as possible. If you have lots of farms (and plenty of living quarters for plenty of people), you won't need to tend farms and waste man power. This is a large percentage of your problem. Realize that if you have plenty of farms, it doesn't matter what food calamity hits, your people will be fine. FYI, morale is easy enough by scavenging frequently. One will often get happiness by scavenging, assuming that you've killed the zombies on these squares. If you're busy collecting two food as long as possible, not only does it add to the likeliness of happiness, such as multiple squares helps provide you needed food, additionally it means that there are fewer zombies that are able to attack your town. As for the 10 day difficulty, the more people you have, the easier it will be to survive. If you have any questions, feel free to pm me.
My internet was down most of yesterday, so I wasn't able to be here to answer questions. The first couple of levels are the most difficult as you don't have your optimum skills as yet, but the game does get easier. If you like strategy games, this one is worth the effort. Banthallus: No, unlike Monster's Den The Book of Dread, there is no Emporium in this original version. So one simply moves any unwanted items to the red X (garbage). Although it is nice to be able to purchase wanted merchandise, this does make it easier to hold onto wanted back ups for different options of how to play one's characters (or even for nice backups on party defeat if you are playing on standard difficulty). Icthaylius: To use a potion, simply drag it to the person whom you wish to use it on. Since this isn't working for you, click menu, then refresh your game and reenter. This should cause everything to work correctly again. Hopefully this solves your problem. Good luck!

The following information Sionyx provided me (but I have never tested the results). I was told that even if you do allow the aforementioned, that there ARE precautions to hopefully keep your fort from committing mass suicide (but it isn't a guarantee). They are: d on't give food for the Day of the Dead; and don't add the Big Graveyard to the fort. Also mentioned is that there is an event where some of your survivors will come to the leader and ask to ban the Church because they're worshiping zombies. If you say yes you will lose the leader of the Church and two other random survivors but the fort will be saved from this ending. If you say no, some of your survivors will leave in disgust and you are well on your way to accomplishing that ending. Elsewhere, I read that when you get a third church that the process is also irreversible. So try to either not get a third church or convert your churches into something else first (such as apartments). Good luck!
Royce: Purchasing items is not what determines how good a game is. That is just what makes a game easier if you don't have the ability (or patience) to play the game without doing so. This game does not bother me with purchasing items. Yes, I can go to the area to do so, but the game doesn't intrude. I never purchase anything, which means that I actually need to use my brain to solve the problems. Of course, I make it even more difficult by not allowing myself to lose one bloon (or I have to start over). This means that I learned well how to improve my game play. You should try it some time. If one likes TDs, Bloons TD games are quite good. My favorite is the fourth one (followed by the third, then the fifth).
Louexveeeye: actually, party defeat depends on which difficulty you select. These are the choices:
Beginner: Monsters have reduced health and damage. No penalty for party defeat.
Standard: Monsters have normal health and damage. Item loss on party defeat.
Hardcore: Monsters have normal health and damage. Game over on party defeat.
Extreme: Monsters have increased health and damage. Game over on party defeat. Fewer loot items.
So you must have played on either Hardcore or Extreme. With the exception of the first few levels, if you lost on Hardcore, then you either don't know how to play your selection of characters well enough or you haven't chosen the right group for yourself. So adjust accordingly to your needs. If you have questions about optimizing any characters by how to use their skills, we can help. Good luck!
RedPandaF0x: You will need to learn which levels present which important new bloons so that you know when you need to place things such as bomb type towers down. Just remember, that for the first one, if you aren’t ready that there are other options. For example, you can use pineapples for your first lead bloons and you can use one set of road spikes for that first camo bloon. But it is very much wasted money if you aren’t prepared for the next time they appear. If necessary, do use those road spikes near the end to prevent bloons from escaping and losing lives. If I know that a round is more difficult than my towers are prepared for, I use glue at such areas as early in the map and where towers are congregated, such as around that first u-turn. Although you don’t want to overuse it, as then you can’t earn enough money, it can help you get through levels as it is definitely cheaper than other options.
